Temeljna je djelatnost udruge Radio Marija proizvodnja i emitiranje radijskoga programa.
Udruga je, sredinom godine 1996., dobila radijsku koncesiju za grad Zagreb. Prvi je studio izgrađen u potkrovlju župne crkve Rođenja Isusova na Kajzerici u Novom Zagrebu.
Veliku potporu projektu Radio Marije dali su blagopokojni Franjo kardinal Kuharić, tadašnji zagrebački nadbiskup, i nadbiskup Giulio Einaudi, prvi apostolski nuncij u Republici Hrvatskoj.
Udruga Radio Marija upisana je u Registar udruga, a ima pravo djelovati na cjelokupnom području Republike Hrvatske.
Sjedište Udruge i sadašnji studio nalaze se na Jordanovcu 110, u župi Bezgrešnog Srca Marijina u Zagrebu.
